Surface sterilization and culturing
See Bascom-Slack et al., 2012, particularly the supplemental material. This formed the basis for our protocol and data sheet (see Appendix). Briefly, leaf tissue is surface-sterilized in ethanol and commercial bleach followed by incubation on appropriate media (PDA or MEA).
Purification and long term storage
We have followed established protocol to purify fungi to generate long-term stocks for later analysis (Bascom-Slack et al., 2012 and A. Fatsi – see Appendix).
